The development of orally active insulin has great benefits to diabetic patient that should administered insulin by injection methods. In order to enhancing the absorption of insulin with stability, we developed a synthetic Nα-deoxycholyl-L-lysyl-methylester and it was complexed with insulin for oral delivery. To make a diabetic condition, we removed all pancreas in dogs via surgical operation and developed a pacreatectomized diabetic dog. Pancreatectomized dogs orally received single doses of insulin (81 U/kg) or insulin/DCK complexes (21, 42, and 81 U/kg) under fasting conditions. The administration of 21, 42 and 81 U/kg (based on insulin activity) of insulin/DCK complexes reduced plasma glucose levels by up to 28.3%, 44.3% and 67%, respectively, compared with baseline levels. The results of this study demonstrate that the insulin/DCK formulation can be absorbed in the intestine. |
