Polyethyleneimnie (PEI) has amine groups that can be combined with DNA. In this report, the polydiacethylene (PCDA) liposomes are prepared and further conjugated with low molecular weight PEI. PEI-grafted PDA liposomes showed good DNA condensation capability and transfection efficiency. It could keep transfection efficiency of PEI and lower toxicity compared to PEI 25kD. So, the PCDA liposome conjugated with PEI was tested as a liposomal vector for gene delivery. We confirmed the complex formation of the polymeric liposome with DNA and then performed transfection and cytotoxicity assay. As the results, these cationic liposomal carriers showed significant gene transfection efficiency and low cytotoxicity. |
