Molecular weight of PEG-PA determines β-sheet to α-helix transition |

Conventional amphiphilic block copolymers tend to self-assemble in water by random hydrophobically driven aggregation, whereas the secondary structural folding motifs such as α-helix and β-sheet affect the assembling process of the polypeptide-based polymers. Poly(L-alanine)10 and PEGylated poly(L-alanine)10 were used as a model system. L-polyalanine with a parallel β-sheeted structure transformed into a antiparallel structure and an α-helical structure in water, and the self-assembled morphology changed into a nanofibrous structure and a spherical micellar structure as the molecular weight of conjugated PEG increased. The sol-gel transition temperature did not follow a simple rule that more hydrophobic polymer has a lower transition temperature, suggesting that the nanoassembled pattern affects the transition temperature. The control of PEG molecular weight is very important not only in designing a PEG-polypeptide biomaterial but also in PEGylation of protein drug. |
