We designed polyamidoamine (PAMAM) dendrimer modified by nuclear localization signal (NLS) peptides. Polyamidoamine (PAMAM) dendrimer is one of the commonly used cationic polymers for gene delivery in non-viral vector system. Because PAMAM dendrimer is consisted of biodegradable chemical bonds, it indicates outstanding biocompatibility and low cytotoxicity. However, the gene transfection efficiency of PAMAM dendrimer shows a generation-dependent aspect and lower transfection capacity compared with that of polyethylenimine (PEI, 25kDa). In this study, we used KKKRK sequence originated from Vp3 of simian virus 40. The KKKRK sequences are introduced at the peripheral ends of PAMAM dendrimer. We performed experiments based on hypothesis that introduction of NLS peptides on the surface of PAMAM may facilitate gene transfer into the nucleus. |
