Hypoxic condition is generally expressed in stroke, inflammation and most solid tumors. Most hypoxic regions are located far from blood vessels(150 um), which provides the conditions of low oxygen level and abnormal tumor tissue environment. In hypoxic condition, solid tumors promote reductive processes and stimuli-sensitive materials utilizing the bioreductive conditions can be applied for activatable sensing systems. In this study, we synthesize a self-immolative dendrimer to apply fluorescence imaging. Dendrimers are expected to display advantages in accessing hypoxic region based on the relatively small hydrodynamic volumes. Nitro group was applied at dendrimers to provide hypoxia sensitivity. After nitro group was reduced in hypoxic condition, self-immolative reaction was sequentially progressed through carbamate linkage. Such changes resulted in the release of fluorescence dyes conjugated at the periphery of dendrimers and produced a hypoxia-sensitive fluorescence imaging probe. |
