제목 | Redox-responsive nanophotosensitizers of β-cyclodextrin-chlorin e6 conjugates via disulfide linkage for photodynamic therapy of cancer cells |
초록 | Photodynamic therapy(PDT) have been investigated for treatment of cancer since PDT using photosensitizers(PSs) generate reactive oxygen species(ROS) in the specific site of light irradiation. Traditional PSs are freely dispersed to the whole body because they have low tumor-specificity. Ce6 was conjugated with succinyl-β-cyclodextrin(bCD) via cystamine linkage(bCDssCe6). bCDssCe6 conjugates formed spherical nanoparticles having small diameter less than 200 nm in aqueous solution. bCDssCe6 nano-PSs significantly increase aqueous solubility of Ce6. Ce6 release from bCDssCe6 nano-PSs was accelerated by addition of glutathione, indicating that they have redox-sensitivity. Furthermore, Ce6 uptake ratio, ROS generation and phototoxicity of bCDssCe6 nano-PSs against HeLa human cervical cancer cells were increased compared to Ce6.
