Amphiphilic block copolymers assemble into nanoscopic core-shell structures, micelles, which are of considerable interest for the delivery of hydrophobic anticancer drug. Targeted drug delivery system is highly desirable to enhance drug efficacy with reducing side effects. We designed polymeric micelles with Poly(ethylene glycol) and Poly(ε-caprolactone) based copolymer containing functional groups between two block segments for drug delivery. PEG block is sterically stabilized and undergo less opsonization and uptake by the macrophages of the reticuloendothelial system, allowing the micelles to circulate longer in blood. Semi-crystalline part of PCL block is biodegradable and resorbable in a suitable period of time. Functional groups are thiol residues which form disulfide bond that lead to stimuli-sensitive release of anticancer drug. |
