Recognition of Hepatocyte in Langmuir-Blodgett Surface of Poly(L-lactic acid)/Poly(ethylene glycol) Diblock Copolymer Endcapped with Galactose Moiety |

The incorporation of galactose ligands into a Langmuir-Blodgett (LB) surface provides the opportunity to develop unique biomimetic surfaces with well-oriented interfaces. Poly (L-lactic acid) (PLA) / poly (ethylene glycol) (PEG) diblock copolymer endcapped with galactose moiety was used as a model ligand for the asialoglycoprotein receptors on hepatocytes. Differences in the adhesion and morphology of hepatocytes between LB and cast surfaces of the PLA, PLA/PEG and PLA/PEG galactose (PLA/PEG-G) polymers were compared. Surfaces of PLA/PEG block copolymer became hydrophilic after incorporation of PEG into the PLA homopolymer. The surface of the PLA/PEG-G copolymer was more hydrophobic than that of the PLA/PEG copolymer. Significant hepatocyte attachment of PLA/PEG-G copolymer LB surface was obtained compared to the cast owing to well-oriented galactose moieties of PLA/PEG-G LB surface. From flow cytometry and confocal laser microscope results, PLA/PEG-G nanoparticles can be recognized specifically through receptor-mediated mechanism. Cells adhering to the PLA/PEG-G copolymer LB surface in the presence of EGF showed round shapes and exhibited spheroid formation after 28 h, whereas the cells adhering to the PLA LB and cast surfaces showed spreading shapes. |
