제목 In vivo Tumor-targeted Imaging by Heavy Metal-free Quantum Dots Encapsulated within Human Serum Albumin Nanocapsules
초록 We introduce a shell cross-linked protein-polymer hybrid nano capsule (NC) as an efficient intratumoral delivery vehicle for highly luminescent Cu0.3InS2/ZnS (CIS/ZnS) quantum dots (QDs). CIS/ZnS-loaded NCs are prepared by emulsifying amine-reactive six-armed polyethylene glycol (PEG) in dichloromethane into aqueous solution containing human serum albumin (HSA). The resulting NCs are stable even in the presence of serum and exhibit the luminescence of QDs comparable with that in non-polar organic solvent, indication high dispersion stability of the NCs. Folic acid (FA) is introduced as a target ligand to facilitate intratumoral uptake of CIS/ZnS-loaded HSA/PEG NCs. In several types of cells, the CIS/ZnS-loaded HSA/PEG NCs show reduced QD-induced cell death compared with cadmium selenide-cored QDs. The in vivo distribution of the CIS/ZnS QDs was monitored for 24 hours after tail vein injection into mice.
