Inhibition of tumor growth by 5-fluorouracil and hydrogel as drug depot |

While administrating anticancer drug for treatment, normal tissue can be affected by dose limiting problem. So, we prepared biodegradable MPEG-b-(PCL-ran-PLLA)(MCL) hydrogel and measured viscosity for the evaluation of thermo-sensitive property. In vivo antitumor activity was performed with 5-fluorouracil(5-FU)-loaded MCL, single and repeated intratumoral injection of 5-FU solution in tumor. The remaining amount of 5-FU drug was measured using HPLC after the removal of tumor from each mouse. For cytotoxicity test WST-1 assay was performed where as for histological analysis H&E and TUNEL processes were performed respectively. The findings in this experiment showed that 5-FU-loaded MCL can act as a biodegradable drug depot. Finally, this work indicates that intratumoral injection of 5-FU-loaded MCL allowed 5-FU to act effectively in the tumor and induced long-lasting inhibition of tumor growth. |
