제목 SPR Sensor for Evaluation of Protein Multivalent Binding
초록 Identification of protein-protein interactions is critical for better understanding of biochemical pathways in biology and medicine. For example, cell signalling by epidermal growth factor is triggered by the receptor dimerization through protein multivalent binding(PMB). Surface plasmon resonance(SPR) sensing has emerged as one of the most reliable platforms for detecting protein interactions with other molecules due to its real-time and label-free manner. However, distinction of PMB is still challenging by using current SPR techniques. Here, we report a bio-responsive microgel-based SPR sensor for the determination of multivalent binding and kinetics. By incorporating bio-responsive microgels to the SPR instrument, we were able to evaluate PMB events. Our results show PD-L1 antibody interaction, being currently used for cancer immunotherapy, is detectable to the level of 10 nM. Our biosensor can also be implemented for the detection of various multivalent species over monovalent one.
