MPEG-PCL diblock copolymer was prepared by the ring-opening polymerization poly(ε-caprolactone) (PCL) with methoxy polyethylene glycol (MPEG, 750 g/mol) as a initiator. Afterward, Gly-Phe-Hyp-Gly-Glu-Arg (GFOGER) was conjugated to the hydroxyl terminal group of MPEG-PCL by the interconnection with succinic anhydride (SA), characterized by 1H NMR (MPEG-PCL-GFOGER). MPEG-PCL hydrogel was formed by dissolving the polymer above a concentration of 20 w/v%, whereas GFOGER-conjugated MPEG-PCL (MPEG-PCL-GFOGER) was not. Rheological measurement revealed that the hydrogels containing GFOGER had two phase transition as temperature was increased from room temperature: a sol-to-gel transition and a gel-to-sol transition. In particular, hydrogels with 2 and 3 wt% of MPEG-PCL-GFOGER were turned into gel phase around body temperature (~37 °C). |
