Tumor-associated macrophages (TAMs) found in close proximity or within tumor tissues express high levels of scavenger receptor-A (SR-A). In this study, as theranotic nanoagents for photodynamic cancer therapy, we synthesized dextran sulfate-chlorin e6 (DS-Ce6) which target SR-A on TAMs. DS-Ce6 was more selectively taken by LPS- and IL-4-treated macrophages via a SR-A mediated endocytosis. Also, DS-Ce6 was more specifically taken up by macrophages, but not by 4T1 tumor cells. Under irradiation, DS-Ce6 notably caused apoptosis of tumor cells by inducing cleaved caspase-3 and PARP levels. In vivo study demonstrated that DS-Ce6 with laser irradiation showed more significant tumor growth inhibition than free Ce6 and DS-Ce6 without laser irradiation. Therefore, TAMs-targeted theranostic nanoagents will have a good potential for photodynamic cancer therapy. |
