Liquid biopsy has been arised to substitute tissue biopsy which has several significant drawbacks. microRNAs (miRNAs) were used as the biomarkers for various diseases including cancers, but conventional diagnostics suffered from high cost, false-positive results, or low sensitivity. Here, we developed the signal amplification method for multiplexed miRNA diagnosis method using hydrogel microparticles by applying a hybridization chain reaction (HCR) to previous hydrogel microparticle-based assay. So that, we succeed to amplify fluorescent signal intensity and thus sensitivity by dozens of times. This approach allows for signal amplification without pre-concentration or amplification of the original target miRNA, which is the primary principle of PCR. We also succeeded to demonstrate the difference between the normal and the patients in specific miRNA profile from urinary exosomal miRNAs which was unavailable previously. |
