We present poly(amino acid)s based polymer nanogels for drug delivery carriers. We developed the cross-linkable amphiphilic graft copolymer platform of poly(succinimide)-PEG (PSI-PEG) copolymers. The reactive PSI can be cross-linked by diamine cross-linkers. Disulfide cross-linked nanogels prepared by cross-linking of PSI-g-PEG micelles using cystamine cross-linkers, and residual PSI units were converted into poly(aspartic acid) (PAsp). The swelling behavior and the structure of nanogels in aqueous solution were analyzed by SANS. Scattering contributions arising from fluctuations of nanogel network are included in the model expression as a Lorenzian function. And Poly core/shell with contant shell thickness model was selected as a form factor model. A positively charged anticancer drug, doxorubicin, was successfully encapsulated within the PAsp core by electrostatic interaction. The disulfide cross-linked nanogels showed disruption of spherical structure at the reductive environment because of the cleavage of disulfide bonds, and the doxorubicin loaded nanogels showed improved anticancer effect. |
