Thermosensitive nanofiber using Pluronic F127 for sustain release of drug |

In this study, thermosensitive poly(ε-caprolactone)/poly(ε-caprolactone)-poly (ethylene glycol-(amine)2) nanofiber [PCL/PCL-PEG NF] conjugated with Pluronic F127 used for loading and release dexamethasone. PCL-PEG block copolymer was mixed with PCL for electrospining by different blending ratio of PCL-PEG from 0% to 100%. And F127 was conjugated to exposed amine group on the PCL/PCL-PEG NF as molar ratio 1:1. In order to characterize Pluronic F127 conjugated PCL/PCL-PEG NF, contact angle, 1H NMR and release test was performed. 1H NMR showed PEO peak at 3.4 ppm in PCL/PCL-PEG-F127, which was not detected in control group. mPEG-FITC micelle was loaded on nanofiber at 4°C and 37°C and released in 1X PBS at 37°C. As the result, more than 5% (w/w) of drug was released out from PCL/PCL-PEG-F127 compared with control for 30days. Therefore, F127 conjugated PCL/PCL-PEG NF will be appropriate device to coat the surface of stent and release drug such as dexamethasone. |
