MMPs-responsive nanofibrous meshes employing MMP-2 siRNA for diabetic ulcer healing |

We prepared a MMP-2 siRNA-loaded nanofibrous matrix for epidermal gene delivery. For controlled release of siRNA loaded in the matrix, a matrix metalloproteinases (MMPs)-cleavable linker was introduced between cationic polymer, linear poly (ethyleneimine) [LPEI] and the matrix so that external MMPs can cleave the linker to release LPEI and siRNA. Many clinical investigations have shown that diabetic patients express higher concentrations of MMPs, including MMP-2 and MMP-9, in foot ulcers, thus leading to an attenuated wound recovery process. Therefore, exposure to higher levels of MMPs during the diabetic wound healing process accelerated release of the LPEI/siRNA complex from the nanofibrous matrix. Upon suppression of MMP-2 by the released MMP-2 siRNA, wound healing rates are expected to be improved in diabetic wounds. |
