Epidermis-dermis junction (DEJ) between the epidermal layer and the dermis layer of the human skin enhances the structural integrity of the skin and activates internal physiological functions. In this study, we present a full-thickness reconstructed skin equivalents with DEJ. We modeled the shrinkage and expansion of responsive hydrogel using expansion ratio of hydrogel with structural ratio in a complicated hydrogel exposed to external stimuli for DEJ formulation on dermis layer. Then, we fabricated three-dimensional DEJ on the dermis layer using programed hydrogel reverse mold. In addition, the formation of the epidermis layer due to the introduction of human epidermal keratinocytes on the dermis layer resulted a full-thickness reconstructed skin equivalents. Overall, this new Reconstructed Skin would be actively used in tissue engineering, drug delivery, and cosmetics assessments. |
