Mosquito-borne fever virus is becoming a serious problem in the world. For effective prevention of it, simple, portable, and cost-effective diagnostic tools in early stage are strongly required. Here, we demonstrate paper-based all-in-one nucleic acid testing system. Viral RNA in serum sample could be simply purified by paper microfluidics. Target RNA is transferred to reaction pad and highly sensitive RT-LAMP amplification is performed following each target primer. During the amplification process, fluorescence signals are generated and can be detected within one hour. Three targets, zika, dengue, and chikungunya, in serum were simultaneously detected on the paper device within 60 min at 65℃. In analysis graph of time-dependent signal, the all-in-one device has ability for quantitative analysis in 5 – 5000 copies of zika RNA. This all-in-one device can be successfully worked from 10 to 50 uL volume of serum samples and clinical species of zika & dengue virus. |
