We investigated growth kinetics of microalgae, Chlorella sorokiniana in microdroplets consist of TAP-medium. Using microfluidic flow-focusing device with high aspect-ratio microchannel, we can evenly distribute microalgal cells and encapsulate single-microalgal cell into microdroplets. To monitor growth of single-microalgal cell, individual microdrops were kept for a week. When single-microalgal cell was in stationary growth phase, we provided microdroplets consist of TAP-medium deprived of nitrogen source to exchange the existing microdroplets consist of TAP-medium. In this way, we can induce vegetative microalgal cells to accumulate lipids. After 4 days, microdoplets which contain lipid-accumulated microalgal cell were moved toward immobilized arrays to analyze their lipid contents. By nile red staining for lipids, we can compare lipid contents of microalgal cells and screen microalgal strains containing high-lipid contents through microfluidic passive valve system. |
