제목 Control of Molecular Interactions within Multilayered Films to Induce the Fibrillation of κ-Casein Amyloids
초록 The interdiffusion of nano-objects within Layer-by-Layer (LbL) films has recently received considerable attention due to its unique molecular transport phenomena. In particular, the use of weak polyelectrolytes (PE) enables us to construct films that are responsive to external pH conditions. In this study, we have established a novel platform based on the fibrillation of proteins within polymeric thin films through the interdiffusion of precursors of amyloid fibrils. κ-Casein and poly(acrylic acid) (PAA) were used as an amyloid precursor and a weak PE to construct LbL films. With the isoelectric point of κ-casein around pH 4.5, the charge density of κ-casein is strongly dependent on pH value. The films (κ-caseins /PAA)9.5 were initially constructed at pH 3.0 and the films were then subject to incubation in pH 5.0 at 80 ℃. The pH value altered the molecular interactions between κ-caseins and PAAs within the films such that amyloid fibrils were formed within the multilayer films.
