제목 Controlled release of rhBMP-2 from Heparin-functionalized PLGA nanoparticles
초록 Bone morphogenetic protein-2 (BMP-2) plays a decisive role during bone regeneration and repair as well as during various stages of embryonal development. Owing to its high potential in new bone formation it requires local delivery at the treatment site. For this purpose various controlled delivery systems e.g. collagen sponge, calcium phosphate etc. have been designed but each has limitations in terms of either release or stability of BMP-2.
The existence of heparin-binding sites in BMP-2 has already been demonstrated by a series of biosensor experiments and heparin appears to potentiate the BMP-2 activity [1]. Here, we characterize the release of rhBMP-2 from heparin-functionalized Poly (L-lactide-co-glycolide) (PLGA) nanoparticles system. The systems show a sustained release profile for several weeks without initial burst. Control of release profile by varying the heparin composition in the nanoparticle systems is also being investigated.
