The novel porous gelatin scaffolds were prepared using overrun process and particle-leaching (sucrose) method. During the overrun process, air is injected into the polymer solution below frozen temperature. In particular, the technique did not require any chemicals or chemical reaction. In addition, Poly (acrylic acid) (PAAc)-modified gelatin scaffolds were prepared for further modification of bioactive molecules. Cross-linking of modified gelatin scaffolds was optimized with both N-(3-dimethylaminopropyl)-N′-ethyl-carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS). The scaffold formed by overrun process showed a dual-pore structure due to the injected air bubbles and ice recrystallization. Morphological investigation revealed that overrun-processed scaffolds had closed pore structures. The overrun process combining particle-leaching (sucrose) method produced matrices with interconnected structure between pore and pore by air bubble. Their pore size and size distribution were significantly influenced by the impeller rate and concentration of polymer solution. |
