제목 Targeted Delivery of Paclitaxel by Folate-conjugated Heparin-based Self-assembled Nanoparticle
초록 We synthesized the folate-conjugated heparin/poly(β-benzyl-L-aspartate) (PBLA) amphiphilic copolymers (HPs) with or without a poly(ethlylene glycol) linker as paclitaxel (PTX) carriers. PTX was incorporated into heparin nanoparticles at a high drug loading content by a simple dialysis method. The size of PTX-loaded nanoparticles was in the range of 140-190 nm, which indicated the potential capacity for targeting solid tumor tissues passively by the Enhanced Permeability and Retention (EPR) effect. In vitro drug release test revealed that these PTX-loaded nanoparticles displayed sustained drug release patterns. Confocal image analysis revealed that folate-conjugated nanoparticles exhibited far greater extent of cellular uptake than the nanoparticles without folate ligands against KB cell. They also showed higher toxicity against tumor cells than PTX itself and folate-unconjugated nanoparticles.
