제목 Mucoadhesive and pH-sensitive thiolated Eudragit microspheres for oral delivery of Pasteurella multocida antigens containing dermonecrotoxin
초록 Oral vaccination is the most attractive route of vaccine delivery. However, low oral bioavailability, degradation and short residence time of antigen at target site are the prime problems. Pasteurella multocida antigens containing dermonecrotoxin (PMD) is a virulence factor of atrophic rhinitis in swine. To overcome these problems, PMD-loaded mucoadhesive and pH-sensitive thiolated Eudragit microspheres (PMD-loaded TEMS) were prepared. Modification of Eudragit® L-100 was done by reaction with L-cysteine HCl. PMD-loaded TEMS were prepared by double emulsion (w/o/w) technique. About 8.9 mol-% of cysteine was introduced to the Eudragit. PMD-loaded TEMS were spherical with smooth surface and average particle sizes were 5.2 m. The TEMS provided higher vaccine loading content and more vaccine was released at pH 7.4 than at pH 1.5. Immune stimulation in Raw 264.7 from PMD-loaded TEMS will be reported. The TEMS will provide a promising delivery system for oral administration of vaccine.
