Multi-functional hydrogel based 3D co-culture system for bone regeneration by paracrine effect |

In this study, paracrine effect between mesenchymal stem cells (MSCs) and osteoblasts was systemically investigated in the collagen-based three-dimensional scaffolds. 3D encapsulation of mesenchymal stem cells in hydrogel scaffolds has been conducted for bone tissue regeneration. Osteoblasts were encapsulated in alginate microbeads using electrostatic droplet extrusion. Osteoblast-encapsulated microbeads were embedded with MSCs in collagen bulk hydrogel scaffolds. We hypothesize that factors secreted from osteoblasts have an influence on osteogenic differentiation of MSCs by the paracrine effect. We prepared three experimental groups which are osteoblasts with MSCs, bone morphogenetic protein 2 (BMP-2) with MSCs and MSCs only to compare the efficiency of paracrine effect. Our system developed in this study can potentially be used to study the paracrine effect from the factors secreted from the cells. |
