제목 Non-invasive optical imaging of the cascade of events during angiogenesis in mouse hindlimb ischemia models
초록 Angiogenesis, which has been an important target for disease diagnosis and therapy, is comprised of a series of events. In the early stage of angiogenesis, the expression of VEGFR and the ECM-deteriorating activities of proteases such as MMP are upregulated. Thus, imaging both events is vital in monitoring the initial process of angiogenesis. Here, we synthesized and characterized two imaging probes which are based on a VEGFR-binding peptide and a MMP-degradable peptide. The probes were used together to visualize and qualitatively measure the VEGFR expression and the MMP activities in mouse hindlimb ischemia models with induced angiogenesis. The combined time-lapse images were employed for the spatio-temporal comparison of the cascade of events during angiogenesis. The non-invasive visualization of the first series of events in angiogenesis indicated that the location of the over-expression of VEGFR changes subsequently following the increased MMP activity in the ischemic region.
