We investigated the growth kinetics of microalgae, Chlorella vulgaris, in microdroplets that consist of TAP-medium. We can evenly distribute microalgal cells and encapsulate single-microalgal cell into microdroplets, using microfluidic flow-focusing device with high aspect-ratio microchannel. To monitor the growth of a single-microalgal cell, individual microdroplets were kept for a week. If we use different single microalgal mutants, there are differences of cell growth among microalgal mutants. To screen microalgal mutants which have high-growth rate, we have to separate the microdroplets encapsulating mutants by density difference using perpendicular flow. The high-density microdroplets go straight to outlet chamber because of the intertial force. |
