제목 Fabrication, Material Characterization and In Vitro Biocompatibility of multi-layered electrospun scaffold for blood vessel tissue engineering
초록 Synthetic polymers have already been well tested as artificial grafts for blood vessels however there still exist problems with regards to proper tissue integration and resulting functionality of these implanted grafts. This study aims to investigate the potential of multi-layered electrospun scaffold for blood vessel tissue engineering by combining thin and thick electrospun fibers into a tubular scaffold. Different fiber diameters was be achieved through modification of the electrospinning parameters of polycaprolactone (PCL). Thin electrospun fibers were used as the inner layer while thick electrospun fibers were used as the outer layer of the tubular scaffold. The inner layer of thin electrospun fibers was prepared to provide a two-dimensional surface for enhanced endothelial cell attachment while the thick outer fiber is intended to produce a three-dimensional environment that would allow cell infiltration. The resulting microarchitecture of the multi-layered scaffold was compared with monolithic thin and thick fiber sacffolds using scanning electron microscopy (SEM). In vitro biocompatibility of the fabricated samples were tested using rat bone marrow stem cells (BMSC). Effect of fiber morphology in terms of macrophage cell infiltration was tested using RAW264.7 cells and endothelial cell attachment behaviour was examined using Cultured Pulmonary Artery Enodthelial (CPAE) cells.
