NONOates-Loaded Block Copolymer Templated Mineralized Nanoparticles for Intracellular Nitric Oxide Delivery |

Nitric oxide (NO) has been known as an important molecule in controlling apoptosis of cancer cells. In these days, mineralized nanoparticles based on block copolymer templates have shown efficient loading potential of therapeutics and delivery ability to target disease sites. In this study, we are heading to develop a NONOates-loaded mineralized nanoparticle (NONOates-MNP) that can intracellularly deliver nitric oxide. The NONOates-MNP was prepared by CaCO3 mineralization using NONOates-incorporated poly(ethylene glycol-b-polyethyleneimine) (PEG-b-PEI) block copolymer as templates. At physiological pH, the CaCO3 core phase of the MNP would effectively protect NONOates. In contrast, at an endosomal acidic pH, NONOates would be exposed to an aqueous environment as CaCO3 core phase degrades, thereby releasing nitric oxide (NO) molecules at the cytoplasm. NONOates-loaded MNP can be used as a novel intracellular NO-releasing nanocarrier for nitric oxide-mediated cancer chemotherapy. |
