제목 Articular Cartilage Tissue Engineering with PLGA/Keratin scaffolds
초록 Keratins are the intermediate filament proteins that form a dense meshwork of filaments throughout the cytoplasm of epithelial cells. We prepared PLGA and PLGA/Keratin scaffolds using solvent casting/salt leaching method. Chondrocytes were isolated from the articular cartilage of rabbit and cultured. Cell proliferation activity was by MTT assay. Morphological observation, histology, biological assay for collagen and sGAG, and PCR were performed at 1, 2 and 3 weeks. The cell viability and the quantity of collagen and sGAG were better PLGA/Keratin scaffolds than PLGA scaffolds. Specific mRNA, collagen type II and aggrecan, for chondrocyte expressed significantly highly in PLGA/keratin scaffold while dedifferentiation marker, collagen type X, did not appear. This study suggests that PLGA/Keratin scaffold may serve as a potential cell delivery vehicle and a structural basis for in vitro tissue engineered articular cartilage.
