Preparation of Novel Photo-labile Linker and Its Application to Solid-phase Peptide Synthesis |

The role of linker is very important in solid-phase peptide synthesis (SPPS). Its cleavage condition must be orthogonal to the synthetic conditions of peptides. Photo-labile linkers are cleaved under certain photonic conditions and make themselves very useful tools in on-bead enzyme assay because of their orthogonal performance to both of acid-labile and base-labile condition. But the existing photo-labile linkers have some weaknesses such as difficulty in synthesis or poor kinetic performance.Herein, we prepared N-Fmoc-3-amino-3-(3,4-dimethoxy-6-nitrophenyl)propionic acid from 6-veratraldehyde in 6 steps in high yield and analyzed its photo-cleavage condition and kinetic performance. |
