Synthesis of Curcumin Derivatives for β–amyloid Plaques Optical Imaging |

Curcumin is a natural product with a variety of biological potential and high photoluminescenes. The extracellular deposition of β–amyloid (Aβ) is considered to be a major pathological property of Alzheimer’s disease(AD) brain and the most significant target for molecular imaging. A visualization of cerebral β-amyloid plaques is the most feasible way to diagnose AD. Recently, it has been suggested that the Alzheimer’s peptide, Aβ, accumulates in the eye. In particular, the rigid conjugated planarity compounds fit into hydrophobic binding channels of Aβ plaques easily. In this study, PEGylated curcumin derivatives were synthesized for binding to Aβ plaques with high photoluminescence and blood circulation. In comparison with Thioflavin S, dye used for histology staining of amyloid, our synthesized PEGylated curcumin derivatives showed high intensity in APP/PS1 mice (AD model) ex vivo staining. |
