제목 | Morphological observation of prolonged exposure time of self-microemulsified BCNU released from PLGA wafer against 9L gliosarcoma cells in vitro |
초록 | 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) has been used as a chemotherapeutic agent for brain tumor but it has very short half-life of 20 min in vitro and less than 15 min in vivo by hydrolysis, Therefore, the exposure of intact BCNU from matrox is very brief and penetration depth (2 - 3 mm) is very short in brain tissue. In the precious study, we enhanced the stability of BCNU in vitro by using self-emulsifying drug delivery systems (SMEDDS) consisted of oil and surfactand and/or cosurfactant. The object of this study is to observe the morphology of 9L glicosarcoma cells according to distance from treated wafer in petridish and prolonged exposure time of SME-BCNU. The self-microemulsified BCNU was incorporated into PLGA wafer as a new polymeric implant. Harvested cells were plated in petridish (2×105 cell/㎖). After 24h, intact BCNU-loaded PLGA wafer and self-microemulsified BCNU-loaded PLGA wafer was placed in contact with the attached cells, respectively. After incubation for specific time period, the cytotoxicity was evaluated by using sulforhodamine B assay (Abs, 520㎚). The cells were observed by microscopy system and photographed by microscope attached camera for specific time period. Cytotoxicity of self-microemulsified BCNU/PLGA wafer against 9L gliosarcoma cells was higher than that of intact BCNU wafer and it was confirmed that SME-BCNU-loaded PLGA wafer inhibited the cell growth more than intact BCNU-loaded PLGA wafer in long distance area from the wafers by observing cell morphology.
