제목 | Controlled Protein Delivery Based on Biodegradable Thermo-Sensitive MPEG-PCL Diblock Copolymer Hydrogels |
초록 | As a number of therapeutic protein drugs became available, the development of suitable protein drug release systems using biocompatible and biodegradable polymers has been focused on various biomedical applications.1 Recently, biodegradable thermo-sensitive hydrogels has many special characters such as good solubility in water, easy drug loading, non-toxicity, and biodegradability. Recently PEG-based diblock and triblock polyester copolymers were studied as injectable drug delivery system via stimulating to temperature.2 To prepare thermo-sensitive sol-gel polymers, we synthesized the diblock copolymer consisting of methoxy polyethylene glycol (MPEG, Mn=750 g/mole) and ε-caprolactone (ε-CL) by ring opening polymerization in the presence of HCl․Et2O at room temperature. (Table 1) The aqueous solution of MPEG-PCL diblock copolymers changed from sol to gel phase as the temperature increased. (Figure 1) Also, we fabricated 20 wt% PBS solutions loading 0.1 % bovine serum albumin, dextran, and insulin for investigating protein release behavior. (Figure 2) In conclusion, the aqueous solution of MPEG-PCL diblock copolymers exhibited the controlled release profiles of BSA, Dextran, and Insulin according as polymer concentration and drug loading amounts.
