Synthesis and screening of biodegradable ionizable lipids for the delivery of nucleic acid drugs. |

Lipid nanoparticles (LNP) formulations based on ionizable lipids have shown great promise for the delivery of nucleic acid drugs. Their delivery efficacy has been well verified, however, challenges associated with cellular and immune toxicity still remain. To improve these limitations, we developed new biodegradable ionizable lipids which include reduction-sensitive linker, disulfide bond. By incorporating glutathione(GSH)-cleavable site via SPDP onto the lipids, it can be reduced and cleaved in the cytoplasm and eliminated more rapidly than non-degradable lipids. Based on the lipids we have synthesized, we performed comparative analysis of these materials in vitro and in vivo to show that the biodegradable LNP have similarly potent gene-silencing activity as compared to the non-degradable LNP while lowering the toxicity issues. |
