Probing molecular interaction mechanism of M13 bacteriophage |

M13 bacteriophages can provide a fascinating and versatile platform for nanobiotechnology due to their unique biological and physicochemical properties. We measured the molecular interaction energies between the genetically engineered m13 bacteriophage and five different functional terminated alkanethiol self-assembled monolayers by using a surface forces apparatus (SFA) which has been used to measure the interaction force between materials directly by contacting the macroscopic surfaces without damaging the surfaces. Effects of pH environments and contact times were also investigated. We verified that the phage tends to strong adhesion energy in acidic condition mainly due to π-π stacking and hydrophobic interaction. However, the interaction energies formed by hydrogen bonding was relatively weak. Therefore, by providing fundamental database of the interaction energies using SFA, it will be used to enhance the performance of phage-based system and adduce future phage applications. |
