Docetaxel (DTX) is one of the most widely used antitumor drugs. However, due to its strong hydrophobicity, DTX shows poor aqueous solubility and safety profile, which results in the reduction of its efficacy. To solve the problems, amphiphilic poly(organophosphazene)-DTX conjugate was synthesized by covalent linkage between carboxylic acid in poly(organophosphazene) and hydroxyl group in DTX. Aqueous solution of the Polymer-DTX conjugate showed a sol-to-gel transition as a function of temperature. In vitro release profile of Polymer-DTX conjugate and in vitro its antitumor effect against human breast, gastric, and prostate cancer cell lines were investigated. Polymer-DTX conjugate inhibited tumor growth more effective and longer than DTX alone in body after local injection at the tumor site. These results suggest the possibility of polymer-DTX as an effective carrier for DTX, but the additional studies will be required. |
