Gelatin methacrylate-based(GelMA) hydrogel has been demonstrated as inexpensive, photopolymerizable, cell responsive hydrogel. Furthermore, previous studies have reported that GelMA-based hydrogels supported proliferation, elongation, and migration of endothelial cells. In this study, we fabricated the injectable GelMA-based microspheres for human endothelial cell deliveries. Micro-sized gelatin spheres were fabricated via electrospraying of GelMA and alginate mixtures (in varying ratios) on CaCl2 solution. By removing the alginate through chelation and further crosslinking of GelMA via UV light, the roughness and porosity of GelMA-microspheres were turned. GelMA-based microspheres improved the angiogenic potential of human umbilical vein endothelial cells (HUVEC) engineered to express hVEGF. Furthermore, injection of GelMA-microspheres carrying hVEGF-secreting HUVECs effectively attenuated hindlimb ischemia. |
