Bioinformatics-inspired protein engineering, functional expression and nano-immobilization of Mugil cephalus epoxide hydrolase for asymmetric resolution of racemic epoxides |
초록 |
The epoxide hydrolase (EH) gene of Mugil cephalus (McEH) was engineered based on bioinformatics. The enantioselective hydrolytic activity was enhanced more than thirty times by site-directed mutagenesis. The mutated gene was efficiently expressed in Escherichia coli in the presence of various types of chaperone. The portion of the soluble fraction increased and the hydrolytic activity was further enhanced more than 50% with coexpressing the pGro7 plasmid. The highly active McEH enzyme was efficiently immobilized onto nanoparticles anchoring Ni2+ ions. |
