Paclitaxel (PTX) is effective for various cancers, especially ovarian and metastatic breast cancer. In this study, we were prepared PTX-loaded lipid nanospheres (LNS) as an anticancer drug carrier for oral administration. Negatively charged LNS loaded PTX prepared. Mean particle diameter and zeta potential value of PTX-loaded LNS showed approximately 80 nm and -75 mV, respectively. PTX entrapment efficiency was about 70%. The negatively charged surface of LNS was modified with a polycationic polymer, chitosan, due to the instability of PTX-loaded LNS in simulated gastric solution and intestinal solution. After surface modification of PTX loaded LNS with chitosan, effects of chitosan on physicochemical properties of PTX-loaded LNS such as particle size, surface charge, stability, permeability etc. were investigated. Coating of PTX-loaded LNS with biocompatible polymer such as chitosan might be useful enhancement of gastrointestinal stability of poorly water-soluble drugs. |
