Hyaluronic Acid/Calcium Phosphate Hybrid Gene Delivery System improves osteogenic differentiation of Human Mesenchymal Stem Cells |

In tissue engineering, Human mesenchymal stem cells(hMSCs) are known to have a great potential. However, current application of hMSCs in therapy has challenges because it is difficult to control that cell in vivo such as differentiation to a target cell type. One of the major difficulties for genetic modification of hMSCs is the lack of an efficient and safe gene delivery system. A conjugated catechol-derivatized hyaluronic acid(dopa-HA) can stabilize biocompatible calcium phosphate(CaP)-based nanoparticles. This complex is used as a carrier for gene transfection to hMSCs for enhanced differentiation. As HA show a specific interaction with CD44 of bone marrow-derived hMSCs, dopa-HA/CaP is used for significantly higher transfection in hMSCs than branced polyethylenimine (bPEI, MW 25 kDa) with no cytotoxicity. The co-delivery of micro RNA 148b and a plasmid DNA encoding bone morphogenetic protein 2 by dopa-HA/CaP achieved remarkably improved osteogenic differentiation of hMSCs. |
