NP regeneration on PLGA scaffold penetrated SIS gel in vitro |

Small intestinal submucusa(SIS) has been used as biomaterials possessing cytokines and biocompatibility. PLGA has strength physical property and biodegradable. PLGA scaffolds impregnated with SIS gel(PLGA/SIS) were prepared as novel intervertebral disc(IVD) replacement by the technique of tissue engineering. PLGA(90K) scaffolds were made by solvent casting/salt leaching method using NaCl(size:90~600 µm). These scaffolds immersed into 1% SIS gel, lyophilized, and crosslinked. Nucleus pulposus was isolated from the IVD. We confirmed cell viability in scaffold. The degree of collagen synthesis was tested through hydroxyproline assay and the formation of disc tissue was evaluated through histology. We chose porogen size of 250~355 µm according to cell viability and hydroxyproline assay. PLGA/SIS scaffold showed good initial attachment, high collagen synthesis, cell viability, and good expression of proteoglycan and type II collagen.- This work was supported by SCRC(SC3100). |
