To reconstruct critical-sized bone defect, a 3D scaffold with growth factors was often utilized for bone healing. In recent, combination of various growth factors has been widely investigated for advanced new bone formation. It is believed that if the physiological processes in human body during bone regeneration can be imitated using growth factors, it will be a promising strategy for effective bone regeneration. However, many studies were not mimicked the growth factor release pattern occurred in the body when bone regeneration because of absence of proper delivery system for the growth factors. In this study, we selected three growth factors (PDGF-BB, VEGF, and BMP-2) which are major growth factors for bone regeneration. The growth factors were released in three different porous particles (initial, continuous, and delayed release). The morphology, release profiles of each growth factor, cell behaviors, and new bone formation potential in rat calvarial defect were estimated. |
