Construction of expression vectors for production of anthracycline aglycones in Streptomyces coelicolor |

Antracyclines, for example, doxorubicin, nogalamycin and mithramycin have used to treat cancer. Anthracycline anticancer agents show cardiotoxicity which is the reason for limited dose of these drugs and there is a big need to develop new anthracylines with diminished cardiotoxicity. We constructed DNA vectors to produce modified anthracycline aglycons. In the last report, we showed that the 13 genes known to be involved in the biosynthesis of ε-rhodomycinone, a intermediate of the doxorubicin, were reorganized into two vectors, each of them containing apramycin and thiostrepton resistance marker gene respectively. We added the snoaL2 gene involved in biosynthesis of C-1 hydroxylation of nogalmycin aglycon into the vector system to modify the aglycon structure. Furthermore we modified the dpsE gene by site-directed mutagenesis to introduce C-2 hydroxylation of the aglycon. The two vectors introduced into S. coelicolor YU105 and the transformants were analyzed for their metabolites by LC-MS. The produced aglycons would be utilized as a substrate for glycosylation of various deoxysugars. |
