제목 Production of recombinant cellobiose dehydrogenase from Phanerochate chrysosporium for enzymatic fuel cell
초록 Cellobiose dehydrogenases (CDH) structure consists of two distinct domains, flavin adenine dinucleotide (FAD) and cytochrome, which contains dehydrogenase and heme B respectively. Based on the enzyme structure, CDH has been reported with high attention in electrochemical applications such as enzymatic fuel cells (EFC) and biosensors.
In this study, heterologous expression of the cdh from P. chrysosporium KCCM 60256 in Pichia pastoris X-33 was achieved for the reliable and efficient production of the enzyme. The anode and cathode of EFC system were modified with recombinant CDH (PcCDH) and laccase, respectively which were composed of graphite oxides immobilized on the modified Au surface. The reaction conditions of the EFC were a 10mM lactose solution at pH 4.5 in 0.1 M citrate buffer. The maximum power density obtained in this EFC with PcCDH was higher than the previosly results. The successful demonstration of this study suggests the potential development of EFC system based on the application of PcCDH.
