This work reports the preparation of silμica-encapsulated Candida antarctica B (CalB) and demonstration of the esterolysis for p-nitrophenyl butyrate. The silica-encapsulated CalB was prepared by sol-gel reaction to control the thickness and enhance the stability as a function of the concentration of tetraethyl orthosilicate (TEOS). The particle size of silica-encapsulated CalB was 4.7 μm and was confirmed by SEM and particle size analyzer. Moreover, these particles were demonstrated the esterolysis for p-nitrophenyl butyrate as the function of used amounts and various pH. The catalytic parameters of Michaelis-Menten kinetics such as Km, Vmax, and Kcat were calculated. Turnover values of silica-encapsulated CalB to p-nitrophenol from p-nitrophenyl butyrate was 9.54 and the catalytic activity was optimized in pH 7.0 at 35 °C. |
