제목 Co-delivery of Akt1 shRNA and paclitaxel by thermo-sensitive conjugated linoleic acid-coupled poloxamer hydrogel
초록 Akt is considered an exciting target for therapeutics. It is suggested that the reduction of Akt expression may increase the therapeutic efficacy of paclitaxel. In the present study, thermo-sensitive CLA-coupled poloxamer hydrogel was synthesized and applied for the co-delivery of Akt1 shRNA and paclitaxel. Biodegradable poly(β-amino ester) (PAE) composed of low molecular weight polyethylenimine and poly(ethylene glycol) was used as a carrier for Akt1 shRNA. The PAE exhibited high transfection efficiency and low toxicity in MCF-7 and MDA-MB-231 breast cancer cell lines. Akt1 shRNA complexed with PAE exhibited significantly gene silencing efficiency in MCF-7 breast cancer cells. Moreover, reduction of Akt1 expression increased the in vitro chemosensitivity of MCF-7 cells with paclitaxel and increased paclitaxel-induced apoptosis. The shRNA-induced knock-down of Akt could be a novel and potent strategy to sensitize cancer cells to paclitaxel for more efficient chemotherapy.
