Surface-decoration of hydrolyzed PCL nanofibrils with photo-crosslinakble gelatin for 3D cell skin model |

We prepared gelatin hydrogel coated on the surfaces of the PCL nanofibrils for 3D cell cultivation. Methacrylate functionalized gelatin (gelMA) that gelMA was adsorbed on the surfaces of hPCL by a dipping method. The gelMA incorporated nanofibrils was photo crosslinked by photo-initiator and UV light. gelMA was more efficient coated on hPCL than gelatin and the water swelling ratio of gelMA@hPCL was much higher than gelatin@hPCL and hPCL. gelMA@hPCL also showed superior biocompatibility to conventional nanofibrils. Human dermal fibroblast cells were cultured with gelMA@hPCL, gelatin@hPCL and hPCL for 14 days. Cells cultivated with gelMA@hPCL showed higher proliferation, and formed better cell/nanofibrils complex compared with other nanofibrils. Thus, gelMA@hPCL hybrid matrix are expected materials with distinct characteristics of both hydrogels and nanofibrils for 3D cell cultivation. |
