For successful treatment of coronary artery disease, t-PA drugs have been used to remove the thrombus. Since these t-PA drugs have short half life and deficient stability in the blood, they should be administrated excessively, which cause side effects including local hemorrhage. In this study, TNK, a type of t-PA drugs, encapsulated within PLGA particles were prepared to increase its biological stability and duration time. The size of the TNK loaded micro/nano-particles were approximately 2 m and 250 nm, respectively. While the microparticles presented porous structure, the nanoparticles showed smooth spherical shape. The drug loading efficiency were about 80 %. When activity of the TNK released from the particles was examined by fibrin zymography, free TNK showed the decrease in its activity with the lapse of time, while the released TNK maintained its stability for over 7 days. Our results suggest that the TNK loaded particles are promising drug carriers for the thrombolysis. |
